GHRP-2

GHRP-2 (Growth Hormone-Releasing Peptide-2) is a synthetic hexapeptide (D-Ala-D-β-Nal-Ala-Trp-D-Phe-Lys-NH₂) belonging to the growth hormone secretagogue (GHS) family. As a potent, metabolically stable agonist of the ghrelin receptor (GHSR-1a), it powerfully stimulates the pulsatile release of endogenous growth hormone (GH) from the anterior pituitary, bypassing the hypothalamic GHRH pathway.

1.MECHANISM OF ACTION

GHRP-2 exerts its primary effects by binding to and activating the Growth Hormone Secretagogue Receptor 1a (GHSR-1a), also known as the ghrelin receptor, on pituitary somatotrophs and hypothalamic neurons. This is a Gq/11-protein coupled receptor. Activation leads to phospholipase C (PLC) stimulation, inositol trisphosphate (IP₃) and diacylglycerol (DAG) generation, intracellular calcium mobilization, and protein kinase C (PKC) activation.

    2.PRODUCT INDICATIONS

  • Endocrinology & Physiology Research: Investigating the regulation of GH secretion, the GHSR-1a pathway, and the synergistic effects with GHRH.

  • Metabolism & Body Composition Studies: Exploring the effects of stimulated GH on lipolysis, protein synthesis, glucose metabolism, and muscle growth in animal models.

  • Aging Research: Studying GH axis function in models of age-related somatopause.

  • Peptide Pharmacology: Serving as a reference standard GHSR-1a agonist for screening antagonists or comparing the efficacy of new growth hormone secretagogues.

  • HPA Axis Studies: Researching the mechanism of GHSR-1a-mediated stimulation of ACTH and cortisol release.

  • Performance & Recovery Models: Used in preclinical models to study the effects of elevated GH on physical performance, wound healing, and tissue repair.

    3.CLINICAL EFFICACY OF PRODUCT

  • Established Potency: It is one of the most potent synthetic GH secretagogues developed, with proven efficacy in stimulating GH release in various animal models and human cell-based assays.

  • Therapeutic Analogs & Context: The knowledge gained from GHRP-2 research directly contributed to the development of later-generation GHSs. Pralmorelin (GHRP-2) has been approved in some countries (e.g., Japan) as a diagnostic agent for GH deficiency.

  • Research-Driven Insights: Studies using GHRP-2 have been crucial in characterizing the GHSR-1a pathway and understanding the potential benefits and risks (e.g., concomitant cortisol increase) of therapeutic GH stimulation.

  • Regulatory Status: It is not an FDA-approved drug. Its sale and use are strictly for non-clinical, non-human research purposes. Any human efficacy data is historical and relates to its use as an experimental agent in early-phase research.
