Histatin 5 CAS NO.104339-66-4

Histatin 5 is a natural, histidine-rich, cationic antimicrobial peptide consisting of 24 amino acids (DSHAKRHHGYKRKFHEKHHSHRGY). It is one of the major functional peptides derived from the Histatin family, primarily secreted by the parotid and submandibular glands in human saliva. Histatin 5 is renowned for its potent, broad-spectrum antifungal activity, particularly against Candida albicans, and exhibit…

1.MECHANISM OF ACTION
Histatin 5 exerts its primary antimicrobial effects through a multi-step mechanism culminating in membrane disruption and intracellular targeting. Initially, the cationic peptide binds to negatively charged components on the microbial cell surface (e.g., phospholipids in fungal membranes or lipopolysaccharides in bacteria). This electrostatic interaction is followed by peptide translocation across the membrane without massive pore formation, a process distinct from many pore-forming AMPs. Once inside the fungal cell, Histatin 5 targets mitochondria, where it induces non-lytic efflux of cellular ATP and other small molecules, disrupts mitochondrial membrane potential, and triggers the generation of reactive oxygen species (ROS).
2.PRODUCT INDICATIONS
- Antifungal/Antimicrobial Research: Testing in vitrosusceptibility of fungal pathogens (especially Candidaspp.) and bacteria to determine minimum inhibitory concentrations (MICs) and study resistance mechanisms.
- Oral Biology & Dental Research: Investigating its role in oral innate immunity, biofilm formation on teeth and mucosal surfaces, and its interaction with other salivary components.
- Mechanism of Action Studies: Exploring the detailed steps of fungal cell killing, mitochondrial targeting, and intracellular signaling pathways activated in response to Histatin 5 exposure.
- Wound Healing Research: Assessing its potential to promote migration and closure of epithelial cells in models of oral and dermal wound healing.
3.CLINICAL EFFICACY OF PRODUCT
- Proven In Vitro& Preclinical Efficacy: It is highly effective at killing Candida albicansand other oral pathogens in laboratory studies.
- Clinical Relevance & Therapeutic Potential: Its presence in saliva is considered a key component of oral innate immunity, protecting against opportunistic infections like oral candidiasis
- Therapeutic Development Challenges: While highly active, the native peptide is susceptible to proteolytic degradation in physiological environments.
- Current Status: No Histatin 5-based drug is currently on the market.
