LHRH, salmon CAS NO.86073-88-3

<p>LHRH, salmon (also known as sGnRH or salmon Gonadotropin-Releasing Hormone) is a decapeptide neurohormone with the specific amino acid sequence: Trp⁷,Leu⁸-LHRH (pGlu-His-Trp-Ser-Tyr-Gly-Trp-Leu-Pro-Gly-NH₂). This variant differs from mammalian LHRH (GnRH) at positions 7 and 8, where Trp and Leu replace Leu and Arg. It is the native releasing hormone in salmonid fish and exhibits potent gonadotropin-releasing acti…

1.MECHANISM OF ACTION

Salmon LHRH functions by binding and activating the specific GnRH receptors (GnRH-R) present in the pituitary gland of salmon and other teleost fish. Similar to its mammalian counterpart, it acts as a ligand for G-protein coupled receptors (GPCRs), primarily of the Gq/11 type. Upon binding to its cognate receptor on gonadotrope cells, it triggers a signaling cascade involving phospholipase C (PLC) activation, production of inositol trisphosphate (IP₃) and diacylglycerol (DAG), leading to intracellular calcium mobilization and protein kinase C (PKC) activation.

2.PRODUCT INDICATIONS

  • Aquaculture & Fisheries Management: Used to induce ovulation and spermiation in commercially important salmonid species (e.g., salmon, trout) and other teleosts for controlled breeding and hatchery operations.

  • Reproductive Physiology Studies: Investigating the neuroendocrine control of reproduction, puberty, and seasonal breeding cycles in fish models.

  • Comparative Endocrinology: Studying the evolution, structure-function relationships, and receptor specificity of GnRH variants across different vertebrate species.

  • In VivoInduction Experiments: Administering to fish via injection or implantation to study its effects on gonadal development, sex steroid hormone levels, and spawning behavior.

3.CLINICAL EFFICACY OF PRODUCT

  • Proven Efficacy in Aquaculture: It is widely used and regarded as highly effective for inducing final gamete maturation and spawning in various farmed fish species, particularly salmonids. Its use has revolutionized hatchery production by allowing for controlled, out-of-season, and synchronized breeding.

  • Standard Practice: The use of salmon LHRH or its analogs (e.g., D-Ala⁶, Pro⁹-NEt sGnRH, also known as Ovaprim™) is a standard spawning induction protocol in commercial aquaculture worldwide. It reliably leads to high rates of ovulation and spermiation.

  • Regulatory Status: It is a key active ingredient in approved veterinary products for fish breeding. Its efficacy and safety profiles for this purpose are well-documented through decades of practical application.
