[Tyr4] – MBP (1 – 11)

<p>[Tyr4]-MBP (1-11) is a synthetic, modified peptide antigen representing the N-terminal encephalitogenic epitope of mouse myelin basic protein (MBP), with a specific tyrosine substitution at position 4. This high-purity research reagent is a critical tool for exploring the fine specificity of T-cell recognition and serves as a prototype for altered peptide ligands (APLs) in the study of autoimmune diseases, partic…

1.MECHANISM OF ACTION

This peptide functions as an altered peptide ligand (APL) for autoreactive T-cells. The tyrosine substitution at position 4 modifies the peptide's interaction with the MHC class II molecule (I-Au) and, more importantly, its engagement with the T-cell receptor (TCR). This altered interaction can induce a differential TCR signaling cascade, potentially shifting the T-cell response from a pathogenic inflammatory (Th1) phenotype towards a regulatory or anergic state, thereby modulating the autoimmune attack on myelin.

2.PRODUCT INDICATIONS

This product is indicated for advanced research in immunology and neurobiology. It is specifically designed for: studying the principles of TCR recognition and signaling, investigating mechanisms of T-cell tolerance and immune deviation, evaluating the therapeutic potential of APLs in the experimental autoimmune encephalomyelitis (EAE) model, and serving as a key reagent in the development of antigen-specific immunotherapies for autoimmune demyelination.

3.CLINICAL EFFICACY OF PRODUCT

[Tyr4]-MBP (1-11) is a research tool and not used directly in clinical practice. Its "efficacy" is demonstrated in preclinical models, where studies have shown that such APLs can effectively suppress or modify EAE disease progression. This proof-of-concept has been instrumental in validating the APL approach, paving the way for clinical trials of similar modified peptides designed to induce protective immunity or tolerance in patients with MS and other autoimmune conditions.
