Melanocyte Associated Antigen gp 100 (17 – 25)

<p>Melanocyte Associated Antigen gp100 (17-25) is a synthetic peptide epitope corresponding to amino acid residues 17-25 of the human glycoprotein 100 (gp100 or PMEL17). This nonameric peptide is a well-characterized, HLA-A*0201-restricted antigenic determinant and serves as a fundamental research-grade reagent for studying cytotoxic T-lymphocyte (CTL) responses against melanoma and other pigment cell-derived cancer…

1.MECHANISM OF ACTION

This peptide is processed intracellularly and presented on the surface of antigen-presenting cells (APCs) or melanoma tumor cells in complex with HLA-A*0201 molecules. When recognized by specific CD8+ T-cell receptors (TCRs) on circulating CTLs, it triggers T-cell activation, proliferation, and the release of cytolytic granules (perforin, granzymes). This leads to the targeted lysis and elimination of gp100-expressing tumor cells, constituting a core mechanism of tumor-specific adaptive immunity.

2.PRODUCT INDICATIONS

This product is indicated for in vitro and ex vivo research in tumor immunology and immunotherapy development. Key applications include: monitoring and expanding tumor-infiltrating lymphocytes (TILs) or peripheral blood CTLs specific for melanoma, evaluating the immunogenicity of gp100-targeted cancer vaccines, serving as a critical antigen in MHC multimer staining (e.g., tetramers) for T-cell detection, and conducting T-cell activation and functional assays (ELISpot, intracellular cytokine staining) for both basic research and clinical trial immune monitoring.

3.CLINICAL EFFICACY OF PRODUCT

As a research tool, this peptide itself is not a therapeutic. However, its use has been integral to the clinical development of gp100-targeted immunotherapies. The gp100(17-25) epitope has been a central component in numerous clinical trials involving peptide vaccines and adoptive T-cell transfer for metastatic melanoma.
