1.MECHANISM OF ACTION

The biological activity of MSHa is primarily mediated through its high-affinity and selective binding to melanocortin receptors (MC1R and MC4R), a subset of G-protein-coupled receptors (GPCRs). Its modified amino acid structure (Acetyl-D-Lys¹¹ and D-Val¹³) enhances binding affinity to target receptors and improves in vitro/in vivo stability compared to native α-MSH fragments. Upon binding to MC1R on melanocytes, MSHa activates the cAMP/PKA signaling pathway, which phosphorylates the microphthalmia-associated transcription factor (MITF), upregulating tyrosinase expression and promoting melanin synthesis.

2.PRODUCT INDICATIONS

Melanocortin receptor research: Exploration of MC1R/MC4R structure-function relationships, serving as a standard ligand for receptor activation assays and agonist/antagonist screening.

Skin pigmentation research: Investigation of melanin synthesis mechanisms, development of tools for studying hypopigmentation and hyperpigmentation disorders.

Metabolic research: Study of MC4R-mediated energy metabolism and appetite regulation pathways, supporting anti-obesity and metabolic syndrome drug development.

3.CLINICAL EFFICACY OF PRODUCT

MSHa is currently in preclinical research stages, with promising efficacy demonstrated in in vitro and in vivo models focusing on melanocortin receptor-related pathways. In cell-based assays, it has shown significant selective activation of MC1R and MC4R, with enhanced stability compared to unmodified α-MSH fragments—providing a valuable tool for optimizing peptide-based drug candidates.
