1.MECHANISM OF ACTION

Neuropeptide EI (NEI) is a 13-amino acid, C-terminally amidated peptide with conserved activity across human, mouse, and rat species, exerting dual biological effects as a Melanin-Concentrating Hormone (MCH) antagonist and Melanocyte Stimulating Hormone (MSH) agonist. It acts by interacting with two key receptor families: it competitively binds to MCH receptors (MCHR) to block MCH-mediated signaling, while activating melanocortin receptors (MCRs) to trigger MSH-related pathways. Upon receptor activation, it modulates intracellular signaling cascades, including cyclic adenosine monophosphate (cAMP) production and calcium ion homeostasis, to regulate physiological processes.

2.PRODUCT INDICATIONS

Investigation of MCH and MSH receptor signaling pathways, as well as their crosstalk in neuroendocrine regulation and behavioral modulation

Screening and validation of MCHR antagonists and MCR agonists via in vitro assays (receptor binding, cAMP detection, and calcium mobilization assays)

Study of circadian rhythm regulation, particularly the rhythmic secretion of ACTH and its link to neuroendocrine homeostasis using human, mouse, and rat models

Research on behavioral neuroscience, including grooming, locomotion, and stress responses, by targeting NEI-mediated signaling pathways

3.CLINICAL EFFICACY OF PRODUCT

In receptor-based assays, it serves as a reliable positive control and reference standard to evaluate the potency of MCHR/MCR-targeted drug candidates for neuroendocrine and behavioral disorders

In mouse and rat models of circadian rhythm disorders, it aids in elucidating the role of NEI in ACTH rhythmicity, supporting the development of novel therapeutics for sleep-wake cycle disturbances

It facilitates the characterization of melanocortin and MCH pathway interactions, providing insights for developing treatments for obesity, anxiety, and stress-related neurological disorders
