FluM1 A2 (58-66) CAS NO.141368-69-6

<p>FluM1 A2 (58-66) (CAS NO. 141368-69-6) is a synthetic 9-amino acid peptide representing an immunodominant epitope (residues 58-66) derived from the Matrix Protein 1 (M1) of Influenza A virus, specifically restricted by the HLA-A*02:01 molecule. This high-purity antigen is a crucial research tool for studying cytotoxic T-lymphocyte (CTL) responses to influenza virus and for the development and evaluation of T-cell…

1.MECHANISM OF ACTION

This peptide functions as a T-cell receptor (TCR) epitope. After intracellular processing of the viral M1 protein within an antigen-presenting cell (APC), this specific 9-mer sequence is loaded onto the peptide-binding groove of the HLA-A*02:01 class I major histocompatibility complex (MHC) molecule. The peptide-MHC complex is then displayed on the cell surface. When recognized by the complementary TCR on a circulating CD8+ cytotoxic T-lymphocyte (CTL), it triggers CTL activation, proliferation, and the release of cytolytic granules (perforin, granzymes), leading to the destruction of virus-infected host cells presenting this epitope.

2.PRODUCT INDICATIONS

This product is indicated for immunology and virology research, particularly in the context of influenza and T-cell immunity. Key applications include: in vitro stimulation and expansion of influenza-specific CD8+ T-cells from HLA-A2+ donors; monitoring CTL responses in infected or vaccinated individuals using assays like ELISpot, intracellular cytokine staining (ICS), or MHC tetramer staining; evaluating the immunogenicity of influenza vaccine candidates; and as a key reagent in studies of TCR recognition, immune memory, and cross-reactivity against different influenza strains.

3.CLINICAL EFFICACY OF PRODUCT

FluM1 A2 (58-66) is a research-grade antigen and not a therapeutic vaccine itself. Its clinical efficacy is demonstrated through its role as a standardized reagent in immunological assays. It has been instrumental in defining a dominant human CTL response against influenza, contributing significantly to our understanding of cellular immunity to the virus. Research using this peptide has helped validate the importance of CTLs in influenza clearance and immunity, supporting the development of universal influenza vaccine strategies aimed at eliciting robust, cross-reactive T-cell responses against conserved internal viral proteins like M1.
