Biological Activity
Hepcidin‑20 is the shorter, 20‑amino acid form of human hepcidin, a liver‑derived peptide hormone that regulates systemic iron homeostasis by binding to ferroportin and inducing its internalization and degradation, thereby inhibiting cellular iron export. It is used in research on iron metabolism, anemia, and inflammation. Bioactivity can be validated in ferroportin‑internalization or iron‑secretion inhibition assays.
Storage Conditions
Lyophilized powder: Store desiccated at –20 °C or below, protected from light, for long‑term stability.
Reconstituted solution: Aliquot and store at ≤ –20 °C; avoid repeated freeze‑thaw cycles.

1.MECHANISM OF ACTION

The peptide functions as the master regulator of iron efflux by binding with high affinity to ferroportin, the sole known mammalian cellular iron exporter. This binding triggers the internalization, ubiquitination, and subsequent lysosomal degradation of ferroportin, effectively blocking the release of iron from macrophages, hepatocytes, and enterocytes into the bloodstream, thereby lowering plasma iron levels in response to inflammation or iron overload.

2.PRODUCT INDICATIONS

Hepcidin-20 is indicated for in vitro research focused on human iron metabolism, including studies of anemia of chronic disease, hemochromatosis, and iron-refractory iron deficiency anemia. It is used in assays to measure ferroportin internalization, to modulate cellular iron export in cell culture models, and to evaluate the activity of hepcidin mimetics or antagonists in drug discovery pipelines targeting iron-related disorders.

3.CLINICAL EFFICACY OF PRODUCT

While Hepcidin-20 itself is a research reagent and not a therapeutic, it is instrumental in validating the hepcidin-ferroportin axis as a drug target. Insights gained from its use have directly informed the development of hepcidin mimetics and ferroportin inhibitors currently in clinical trials for conditions like β-thalassemia and polycythemia vera, though the peptide salt form has no direct clinical efficacy data in humans.
